No Follow and Do Follow

Can anyone please state the difference between no follow and do follow? How can you tell once you are on a site which one it is?

Nofollow links has rel=nofollow attribute attached to it, when you check in source file.

Simply, you can install NoDofollow addons on your Firefox browser which would tell you nofollow or dofollow links by highlighting with different colors.

Nofollow links is no useful, and the dofollow links make contribute to the pagerank.

To be honest with the issue, I am still confused with the do-follow link and the no-follow link. In one discussion that I had read, it was stated that do-follow URL means it can be beneficial to you and the no-follow URL is not. Now I am wondering what happens when the link is a no-follow, what happens to that? And when it is a do-follow will it be like an ordinary backlink? Pardon my ignorance.
